ABSTRACT

OBJECTIVE: To investigate the effects of bio-crosslinker genipin pretreatment on type Ⅰ collagen mineralization. METHODS: Type Ⅰ collagen gels were prepared and pretreated with 0.5wt%genipin (experimental group) and deionized water (control group) for 2 h, respectively. The pretreated products were subjected to Fourier transform infrared spectroscopy (FT-IR). Reconstituted collagen fibrils were pretreated with genipin or deionized water for 2 h and were mineralized for 4 h. The collagen density and mineralization degree were examined with transmission electron microscopy (TEM) and analyzed with ImageJ software. Then scanning electron microscopy (SEM) and TEM were used to observe the mineralization of cross-linked demineralized dentin collagen. RESULTS FT-IR spectrum showed that the genipin was crosslinked with collagen. TEM observation and ImageJ results showed that after 4 h mineralization, the mineralization effect of 0.5wt% genipin group was significantly better than that of the control group[(73.3±5.3)%vs.(7.4±3.5)%,P<0.01]. TEM and SEM observation showed that the mineralization rate of type Ⅰ collagen and demineralized dentin pretreated with genipin were significantly faster than that of the control group. CONCLUSIONS The study demonstrates that 0.5 wt% concentration of genipin can significantly promote the mineralization of type Ⅰ collagen.

ABSTRACT

OBJECTIVETo determine the influence of altered masticatory loading on Indian hedgehog (Ihh)-parathyroid hormone-like related protein (PThrP) pathway in the condylar cartilage of growing rabbits.

METHODSA total of 48 10-day-old rabbits were randomly divided into two groups and fed different kinds of food, such as solid diet and soft diet. The animals were sacrificed after 2, 4, 6, and 8 weeks. Difference of Ihh and PThrP expression levels induced by altered masticatory loading was tested by hematoxylin-eosin (HE), immunohistochemistry, Western blot, and real-time polymerase chain reaction (PCR).

RESULTSThe thickness of condylar cartilage and expression levels of Ihh and PThrP proteins and mRNA of the solid diet groups exceeded those of the soft diet groups. The decreasing tendencies of the expression levels of Ihh and PThrP proteins and mRNA were observed at 2, 4, 6, 8 weeks.

CONCLUSIONSLow masticatory loading may delay or inhibit the development of condylar cartilage and its growing factors Ihh and PThrP. Therefore, masticatory loading plays an important role in the development of condylar cartilage.

ABSTRACT

Objective@#To investigate the effect of different mechanical loading on the expression of Notch-1, Notch-3, Jagged-1 and Delta-like-1 in growing rabbits' condylar cartilage.@*Methods@#Sixty-four ten-days-old rabbits were randomly divided into experimental and control groups. The rabbits in the experimental groups were fed on a powder diet, while the control groups were fed on a solid diet. The animals were sacrificed after 2, 4, 6 and 8 weeks. Notch-1, Notch-3, Jagged-1 and Delta-like-1 gene and protein expressions were examined by HE, immunohistochemistry, Western blotting and quantitative real-time PCR.@*Results@#At 2, 4, 6, 8 weeks, the anterior part of condylar cartilage in solid diet groups ([318.1±4.3], [342.4±2.6], [364.2±3.2], [380.7±6.0] mm, respectively) were thicker than those in powder diet groups ([275.4±2.6], [301.1±2.0], [322.3±3.3], [366.5±8.4] mm, respectively) (P<0.05). There was no significant difference between the two groups in the middle part (P>0.05). From 2 to 6 weeks, the posterior part of cartilage in solid diet groups ([444.1±1.5], [451.1±0.3], [476.4±5.7] mm, respectively) was thinner than those in power diet groups ([470.4±2.7], [494.3±2.9], [512.3±5.7] mm, respectively) (P<0.05). At 6 weeks, there were more mRNA expressions of Notch-3, Jagged-1 and Delta-like-1 in solid diet group than in power diet group (P<0.05). The Notch-1 protein could be found in all layers of condylar cartilage, especially in the surface of hypertrophic zone. The expressions of Notch-1, Notch-3, and Delta-like-1 protein were increased from 2 to 6 weeks, and decreased at 8 weeks. In solid diet groups, the protein expressions of Notch-1 (at 2, 4, 6, 8 weeks), Notch-3 (at 2, 4, 6 weeks), Jagged-1 (at 2, 4, 6 weeks) and Delta-like-1 (at 4, 6, 8 weeks) were greater than in power diet groups (P<0.05).@*Conclusions@#Low masticatory loading may delay or inhibit the development of condylar cartilage and its growing factors such as Notch-1, Notch-3, Jagged-1 and Delta-like-1. Appropriate masticatory loading plays an important role in normal development of the condyle.

ABSTRACT

OBJECTIVETo examine the expression of proteoglycan 4 (PRG-4) induced by hydrostatic pressure in rat temporomandibular synovial fibroblasts and investigate the possible mechanism.

METHODSThe cultured rat temporomandibular synovial fibroblasts were subjected to 100 kPa magnitude intermittent hydrostatic pressure (IHP) at frequency of 4 h/day, and the static group served as control. The expressions of Smad pathway proteins and p38MAPK pathway proteins were analyzed by Western blot and immunofluorescence staining. Then the cells were incubated with SB431542, the inhibitor of transforming growth factor (TGF)-β receptor. Western blot and reverse transcription PCR were used to detect the PRG-4 expression after 72 h.

RESULTSThe expression of phosphorylated Smad-2 and phosphorylated Smad-3 were increased after 1 h of IHP, reaching a maximum after 2 h and 4 h of IHP, respectively. However, the protein content of phosphorylated p38 did not vary significantly. In addition, IHP induced nuclear translocation of Smad-2/-3, and the immunofluorescence staining signal intensity markedly increased (24.11 ± 4.70)(P < 0.05). The levels of PRG-4 mRNA were significantly increased by IHP (1.48 ± 0.08)(P < 0.05). Treatment of cells with SB431542 could decrease the expression of PRG-4 mRNA significantly after IHP (0.47 ± 0.05)(P < 0.05). In addition, SB431542 inhibited the expression of PRG-4 protein induced by IHP.

CONCLUSIONSSmad signal acts as an essential signal pathway to regulate PRG-4 expression induced by IHP.

ABSTRACT

OBJECTIVETo study the chondrocyte apoptosis in rabbit temporomandibular joint after anterior disc displacement.

METHODSExperimental anterior disc displacement was induced surgically in 20 Japanese rabbits without opening their temporomandibular joint bursas. Histopathologic and apoptotic (TUNEL) analysis was used to evaluate the changes in articular cartilage, disc and synovium.

RESULTSCondyle chondrocyte showed apoptosis most obviously at 1 or 2 weeks after surgery, and apoptotic cells concentrated in proliferative zone and hypertrophic zone. 4 or 6 weeks after surgery, the joint went into a remodelling period.

CONCLUSIONSChondrocyte apoptosis in temporomandibular joint will be activated after anterior disc displacement, which initiates the remodelling in temporomandibular joint.

ABSTRACT

OBJECTIVETo study the adaptive alterations of elastic fibers in the bilaminar zone (BZ) of rabbit temporomandibular joint (TMJ) following disc displacement.

METHODSTwenty-eight Japanese white rabbits were used in this study. The right temporomandibular joints of 20 of 28 rabbits were subjected to the surgical procedure of anterior disc displacement (ADD). Four rabbits in the surgical group were sacrificed at 2, 4, 6, 8 and 12 weeks after operation. Their temporomandibular joints were studied histochemically.

RESULTSElastic fibers were reduced in number and ran irregularly in the superior lamina of BZ from ADD rabbits. The jungly elastic fibers (EFs) could still be seen at 2 weeks after operation. At 4 weeks, the number of EFs decreased significantly; EFs lost their jungly arrangement and were shaped like rough dots, of which the arrangement and the lengths were different. Six weeks after operation, many EFs were replaced by distorted, uneven, non-oriented fine EFs, distributed unevenly and some thick or fine EFs that ran irregularly. The number of EFs decreased further and their arrangement was more deranged at 8 weeks. At 10 and 12 weeks, EFs in the superior lamina of BZ were similar to those at 8 weeks.

CONCLUSIONOur results show that EFs lost their function as well as their distribution and arrangement after disc displacement.

ABSTRACT

The spleno-and thyme-lymphocytes of mice were cultured in the media with various concentrations of zinc. The results showed that zinc acted as a weak mitogen and stimulating lymphocyte proliferation at 0.12-0.5? 10-4 mol/L. It also enhanced the lymphocyte proliferation to Con A and LPS. The optimal concentration was 0.12?10-4mol/L. It enhanced the MLR of spleno-lym-phocyte and increased the responsive ability of thymocyte to IL-1. Zinc was poisonous to cells at concentrations higher than 1.0 ?10-4mol/L.

ABSTRACT

The nutritional survey was made in 184 anorexia children aged 3-7, 123 of them were treated with various measures for three months. The results indicated that the children with anorexia had low intake and deficiency in energy, protein, minerals and vitamins. Most of them had zinc deficiency, but they simultaneously had two to three nutrients deficiency. The zinc treatment could improve most patient's syndrome and nutritional status. The effect of zinc treatment with vitamines and iron preparations was better than zinc treatment only. The effect of treatment with Chinese medicine also was able to cure the children with anorexia to certain extent.
